Are the stats numbers actual humans pressing the "play" button? Is it counted when people click the link to the sermons pages? Or, is there a possibility that some of those numbers are bot or search engine related?

The stats count every time the sermon is requested: either by pressing the play button, clicking on the download link (if there is one), or downloading file from the podcast. Clicks from admins and iTunes feeds aren't counted. Bots could be counted if you have download links showing.
