Do you see the error when you first click on the Options button, or when you try to hit save on the Options screen?

Could you post a screenshot of what your Options screen looks like before you hit Save and get the error?  If not, could you post what is in each box on the Options screen?

mein, I'd like to see your .htaccess file for your WordPress install.  This is the file that is in your main WordPress directory on your web server.

I'm not sure if it is a security risk or not for you to post that file on a public forum, so I've left you a Private Message with my e-mail address.  Please e-mail me your .htaccess file.

You can deactivate, delete, install, and activate the plugin, and all your sermons will still be there.  (Although, backing up your database is always a good idea before making any changes with plugins.)  The thing you DON'T want to do is hit the Uninstall button (Sermons, Uninstall).  If you do that, it will delete all your Sermon Browser data from the database.

I'm leaning toward the problem being some sort of server configuration.  You should try talking to your web host and see if they will help you.

The Options screen doesn't update any files.  The options are saved in the WordPress database.
